Risk Factors Affecting Muscle Mass Decline in Maintenance Hemodialysis Patients

Objective. There is a high prevalence of sarcopenia in maintenance hemodialysis (MHD) patients, which is known to be associated with increased mortality. This study is aimed at analyzing the risk factors affecting muscle mass decline in MHD patients. Methods. This retrospective study included MHD patients who underwent two body composition assessments in October 2013 and November 2017. more » ... oup and a muscle mass decline (MMD) group. According to the muscle mass decline rate, patients in the MMD group were further classified into a low-rate group and a high-rate group. Biochemical variables, serum vitamin concentrations, anthropometric data, SGA, muscle mass, handgrip, and daily steps were assessed. Risk factors for muscle mass decline were screened by multivariate logistic analysis and linear regression analysis. Results. Of the 72 MHD patients included in this study, 33 were male and 39 were female with a mean age of 56.80 ± 10.86 years and a mean dialysis duration of 7.50 ± 5.20 years. Age ( P = .014 ) and serum 25(OH)D ( P = .040 ) were found to be associated with a higher risk of muscle mass decline after adjusting for gender, dialysis vintage, albumin, and hs-CRP ( P = .040 ). Further analysis found that dialysis vintage ( β = 0.285 , P = .030 ), 25(OH)D ( β = − 0.351 , P = .007 ), and log NT-proBNP ( β = 0.312 , P = .020 ) were risk factors associated with the muscle mass decline rate in MHD patients. Conclusion. Age and serum 25(OH)D were associated with a higher risk of muscle mass decline, while 25(OH)D, dialysis vintage, and NT-proBNP were associated with the muscle mass decline rate in MHD patients.

Long noncoding RNAs (lncRNAs) can promote hepatocellular carcinoma (HCC) initiation and progression. In this report, we examined the role of lncRNA LINC02476 in HCC. The expression levels of different lncRNAs in HCC were explored using the TCGA database and lncRNA LINC02476 was selected for further study. more » ... pression of LINC02476 was downregulated in HCC cells using a lentiviral vector and different assays were performed to study cell growth, proliferation, invasion, apoptosis and the cell cycle. MiR-497 was selected as a miRNA that could interact with LINC02476 which was further tested by RNA immunoprecipitation. HMGA2 was selected as a possible target of miR-497, and their interaction was examined by a luciferase reporter assay. LINC02476 expression was elevated in HCC cell lines and HCC tissues. When LINC02476 was downregulated, the growth and the invasion of HCC cells decreased in vitro and in vivo. LINC02476 negatively regulated the expression of miR-497 by acting as a ceRNA. HMGA2 was directly targeted and inhibited by miR-497. The results indicate that LINC02476 functions through the miR-497/HMGA2 axis and that it has a role in the growth and metastasis of HCC cells. Therefore, LINC02476 could be an interesting new molecular target in HCC therapies.

A Practical Electronic Health Record-based Dry Weight Supervision Model for Hemodialysis Patients

Objective: Dry Weight (DW) is a typical hemodialysis (HD) prescription for End-Stage Renal Disease (ESRD) patients. However, an accurate DW assessment is difficult due to the complication of body components and individual variations. Our objective is to model a clinically practicable DW estimator. Method: We proposed a time series-based regression method to evaluate the weight fluctuation of HD patients according to Electronic Health Record (EHR). A total of 34 patients with 5100 HD sessions
more » ... a were selected and partitioned into three groups; in HD-stabilized, HD-intolerant, and near-death. Each group's most recent 150 HD sessions data were adopted to evaluate the proposed model. Results: Within a 0.5 kg absolute error margin, our model achieved 95.44%, 91.95%, and 83.12% post-dialysis weight prediction accuracies for the HD-stabilized, HD-intolerant, and near-death groups, respectively. Within a 1%relative error margin, the proposed method achieved 97.99%, 95.36%, and 66.38% accuracies. For HD-stabilized patients, the Mean Absolute Error (MAE) of the proposed method was 0.17 kg ± 0.04 kg. In the model comparison experiment, the performance test showed that the quality of the proposed model was superior to those of the state-of-the-art models. Conclusion: The outcome of this research indicates that the proposed model could potentially automate the clinical weight management for HD patients. Clinical Impact: This work can aid physicians to monitor and estimate DW. It can also be a health risk indicator for HD patients.

Increasing plant density is one of the most efficient ways of increasing wheat (Triticum aestivum L.) grain production. However, overly dense plant populations have an increased risk of lodging. We examined lignin deposition during wheat stem development and the regulatory effects of plant density using the wheat cultivars shannong23 and weimai8. more » ... nced culm quality, as revealed by increased culm diameter, wall thickness and dry weight per unit length, and improved the structure of sclerenchyma and vascular bundles by increasing lignification. In addition, more lignins were deposited in the secondary cell walls, resulting in strong lodging resistance. The guaiacyl unit was the major component of lignin and there was a higher content of the syringyl unit than that of the hydroxybenzyl unit. Furthermore, we hypothesised that the syringyl unit may correlate with stem stiffness. We describe here, to the best of our knowledge, the systematic study of the mechanism involved in the regulation of stem breaking strength by plant density, particularly the effect of plant density on lignin biosynthesis and its relationship with lodging resistance in wheat.

Comparative Performance of Creatinine-Based GFR Estimation Equations in Exceptional Longevity: The Rugao Longevity and Ageing Study

Reduced kidney function has been associated with an increased risk for adverse outcomes. more » ... als and is an ideal model to address factors relating to life span and age-related diseases. This study used 6 years' data of 278 EL from the Rugao longevity cohort. Baseline GFR was estimated using four equations: Chronic Kidney Disease Epidemiology Collaboration (CKD-EPI) equation, Modification of Diet in Renal Disease Study (MDRD) equation, Berlin Initiative Study-1 (BIS) equation, and modified MDRD equation. Predictive validity was tested using Cox proportional hazards analysis. Overall improvement in reclassification based on estimated GFR (eGFR) was assessed applying net reclassification improvement (NRI). Mean age of participants was 97±2 years with median follow-up of 2.6 years. Median (IQR) eGFR by CKD-EPI, MDRD, BIS, and modified MDRD equations were 73.9 (62.2-77.6), 82.3 (67.4-98.6), 56.4 (47.9-63.9), and 101.5 (83.1-121.6) mL/min per 1.73 m2, respectively. Higher eGFREPI was associated with lower mortality after multivariate adjustment (for continuous eGFREPI, HRtwo-year 1.018, 95% CI 1.002-1.033, P=0.023; HRsix-year 1.013, 95% CI 1.002-1.025, P=0.022), while eGFR from other equations did not show any associations with mortality. NRI for two-year mortality was 0.14 and approximately significant, which may favor the CKD-EPI when compared to BIS equation (P=0.052). The CKD-EPI equation showed more accurate estimation of kidney function in the elderly with respect to GFR distribution and predictability of mortality risk.

HOTAIR rs7958904 polymorphism is associated with increased cervical cancer risk in a Chinese population

Previously, we have identified single nucleotide polymorphisms (SNPs) rs7958904 and rs4759314 in long non-coding RNA HOX transcript antisense RNA (HOTAIR) were significantly associated with risk of colorectal and gastric cancer, respectively. Here, we aimed to investigate the association between HOTAIR SNPs and cervical cancer (CC) susceptibility. more » ... Atlas (TCGA) database was utilized for in vivo analysis of allele-specific HOTAIR expression. MTT assay was employed for evaluation of allele-specific cell proliferation. The rs7958904 CC genotype was related to an increased risk of cervical cancer compared with the GG/GC genotypes (OR = 1.57, 95% CI = 1.10-2.25). TCGA database showed the CC tissues with rs7958904 CC genotype had higher HOTAIR expression than those with GG genotype (P = 0.046). MTT assay demonstrated a growth-promoting role of rs7958904 C allele on CC cells. Further functional studies on the effect of rs7958904 on biological behavior of CC cells are needed to confirm and extend our findings. In conclusion, HOTAIR rs7958904 might influence CC susceptibility through modulation of CC cell proliferation, and could serve as a diagnostic biomarker. Cervical cancer (CC) is one of the most common causes of cancer mortality for women worldwide. According to the International Agency for Research on Cancer (IARC) (GLOBOCAN 2012), an estimated of 266,000 deaths from CC occurred worldwide in 2012, accounting for 7.5% of all female cancer deaths 1 . Although infection with high risk human papillomavirus (HR-HPV) has been accepted as the major risk factor of CC, individual genetic heritability also plays a fundamental role in the development of CC 2-4 . Long noncoding RNAs (lncRNAs) are a kind of non-coding RNAs longer than 200 nucleotides 5, 6 . The ENCODE project suggests that about 76% of the human genome is transcribed to a series of lncRNAs 7 . Plant Density Effect on Grain Number and Weight of Two Winter Wheat Cultivars at Different Spikelet and Grain Positions

In winter wheat, grain development is asynchronous. The grain number and grain weight vary significantly at different spikelet and grain positions among wheat cultivars grown at different plant densities. In this study, two winter wheat (Triticum aestivum L.) cultivars, 'Wen-nong6' and 'Jimai20', were grown under four different plant densities for two seasons, in order to study the effect of plant density on the grain number and grain weight at different spikelet and grain positions. more » ... showed that the effects of spikelet and grain positions on grain weight varied with the grain number of spikelets. In both cultivars, the singlegrain weight of the basal and middle two-grain spikelets was higher at the 2 nd grain position than that at the 1 st grain position, while the opposite occurred in the top two-grain spikelets. In the three-grain spikelets, the distribution of the single-grain weight was different between cultivars. In the four-grain spikelets of Wennong6, the single-grain weight was the highest at the 2 nd grain position, followed by the 1 st , 3 rd , and 4 th grain positions. Regardless of the spikelet and grain positions, the single-grain weight was the highest at the 1 st and 2 nd grain positions and the lowest at the 3 rd and 4 th grain positions. Overall, plant density affected the yield by controlling the seed-setting characteristics of the tiller spike. Therefore, wheat yield can be increased by decreasing the sterile basal and top spikelets and enhancing the grain weight at the 3 rd and 4 th grain positions, while maintaining it at the 1 st and 2 nd grain positions on the spikelet. 3 / 15 **Significant at P < 0.01.
